In-vitro thrombolytic activity of Curcuma longa linn (turmeric) rhizomes extract.

Abstract

      Certain plants have been shown to have significant applications in the medical field. One such aspect is in the field of thromboembolic diseases which have a relatively high mortality rate. Curcuma longa Linn, a herbal plant, commonly used as a spice, was demonstrated to induce clot lysis through its active component curcumin found at the rhizome of Curcuma longa Linn plant. Curcumin was proposed to inhibit two major inhibiting proteins of the fibrinolytic system which are thrombin and Plasminogen Activator Inhibitor-1. This study aimed to confirm and evaluate the clot lysis activity of Curcuma longa Linn in vitro. A crossover design was utilized to evaluate the clot lysis of Curcuma longa Linn, an in vitro approach was done by using the Euglobulin Clot Lysis Time method. This method measures the euglobulin portion of a blood specimen which contains significant proteins involved in clot lysis such as plasminogen. To measure its effectiveness, the clot lysis time of Curcuma longa Linn was compared to that of Streptokinase, a standard thrombolytic agent, and the untreated group. Plant extraction was done via soxhlation. The blood of fifteen healthy donors was used for the experimentation. The results were analyzed using One-way Repeated-Measures ANOVA with Greenhouse-Geisser adjustment and Bonferroni Multiple Comparisons Test.


      One-Way Repeated-Measure ANOVA revealed clot lysis time of the blood samples treated with turmeric rhizomes extract, Streptokinase as positive control, and untreated samples were not the same at .05 level of significance, F(1.14) = 612.160 and p<.05. It was found out that the clot lysis time of blood samples treated with Curcuma longa Linn rhizomes extract (Mean = 2.83, SEM = 15 & 95% Cl [2.50, 3.16]) was significantly longer compared to Streptokinase-treated samples (Mean = 1.37, SEM = .10 & 95% Cl [1.17, 1.58]). However, the rhizomes did possess in-vitro thrombolytic activity for it lysed the clot in less than two hours. In addition, the clot lysis time of the blood samples treated with Curcuma longa Linn rhizomes extract (Mean = 2.83, SEM = 15 & 95% Cl [2.50, 3.16]) was significantly shorter compared with the blood samples in the untreated group by 1,141.85 minutes.


      The results indicate that Curcuma longa Linn does possess the property of clot lysis because it lysed the thrombus in a short span of time which is less than two hours. Further studies should be made to evaluate thrombolytic activity utilizing the different parts of the Curcuma plant.
