Atis (Annona squamosa) leaf extract as an alternative selective media for Escherichia coli growth.

Abstract

          This study determines the potential of Annona squamosa leaves locally known as Atis, as a cost - effective, locally grown and easily obtained alternative culture medium for Escherichia coli growth. The study was conducted both in the Cebu Doctors' University Hospital and at Room 404 of Cebu Doctors' University Medical Technology Laboratory. The test was carried out against two selected microorganisms, namely Escherichia coli and Staphylococcus aureus. The experimental control group was comprised of 144 melted nutrient agar plates incorporated with 100% Annona squamosa leaf extract, while the positive control group was consisted of 144 plates of MacConkey agar. Escherichia coli was inoculated in seventy - two (72) plates from the experimental group and seventy - two (72) plates from the control group. The procedure was repeated with Staphylococcus aureus. The proportion of growth between
the experimental and control group was determined after 24 hours of incubation.
          According to the data that was collected, Escherichia coli showed growth in both the 72 melted nutrient agar plates incorporated with 100% Annona squamosa leaf extract and 72 plates with MacConkey agar. The agars were examined for the characteristic growth of gram-negative Escherichia coli. Further biochemical tests were performed to confirm the presence of Escherichia coli seen on the plates. The results of the experiment were tabulated and the frequency and percentage distribution revealed 100% growth on both experimental and control groups. On the other hand, the plates inoculated with Staphylococcus aureus showed no significant growth in both the 72 melted nutrient agar plates incorporated with 100% Annona squamosa leaf extract and 72 plates with MacConkey agar. The results of the experiment were also tabulated and the frequency and percentage distribution revealed 100% growth inhibition on both experimental and control groups.
          Based on the results of the study, the researchers concluded that the Annona squamosa leaf extract was effective in inhibiting Staphylococcus aureus and promoting the growth of Escherichia coli. Therefore, it can be said that the Annona squamosa leaf extract can be used as an alternative culture medium for Escherichia coli growth.
