Charcoal ink as primary stain in the capsular staining of Klebsiella pneumoniae using hiss method.

   The study aimed to determine the potential of charcoal ink as an alternative primary stain for the visualization of the capsule of the bacterium Klebsiella pneumoniae. The researchers utilized the single blind experimental randomized controlled trial with multiple level of treatments as its research design. This study included ninety (90) slides with the bacterial suspension of Klebsiella pneumoniae. Two sample dilutions were used (1:5 and 1:10 dilutions) and the crystal violet was used as the positive control. Thirty slides were allocated to each of the group. Hiss Staining Method was employed to visualize the organism.

   The findings revealed that the capsule of Klebsiella pneumoniae using the 1:5 and 1:10 dilutions of the charcoal ink was not exhibited for the entire sixty (60) slides. Distorted capsules and significant amount of debris was observed in the microscopic field. However, thirty slides were noted to positively stain the capsule using the crystal violet as the primary stain. The control group using crystal violet positively stained the thirty slides.

   Based on the findings, charcoal ink at dilutions 1:5 and 1:10 has proven to be inefficient in the visualization of the capsule of the bacterium Klebsiella pneumoniae. It is therefore recommended to consider the utilization of the charcoal ink in the staining of gram positive organism without special structure using the Negative Staining method. In this method charcoal ink will be added to the organism after fixing and immediately examined using the microscope.
