Thrombocytosis effect of Carica papaya seed extraction on the platelet count of aspirin-induced albino rabbit (Oryctolagus cuniculus).

Abstract

This study aimed to determine the ability of Carica papaya seed extract to increase the platelet count of aspirin-induced thrombocytopenic albino rabbits. The experimental method used was a randomized controlled trial, single blind. Fifteen (15) male albino rabbits were divided into three (3) groups, composed of five (5) rabbits each. Aspirin was administered to the subjects for a period of one (1) week in order to simulate a thrombocytopenic state. Carica papaya seed extract was obtained by subjecting dried papaya seeds to ethanol extraction with the use of the Soxhlet apparatus and Rotavap. The three (3) groups received different dosages of the Carica papaya extract for fourteen (14) days: experimental group 1 received 0.5 mL/kg of seedextract, experimental group 2 received 1 mL/kg of the extract, and the negative control group did not receive any extract, only distilled water. Indirect platelet counting was done during the 7th and 14th days.


The findings of the study demonstrated a significant difference across time in all three groups. Using Repeated Measures ANOVA, it was found that for the 0.5 mL/kg Carica papaya group, aspirin-induced platelet count was significantly lower compared with day 7 post-treatment by 207,345 plt/μL, but day 7 and day 14 post-treatment platelet counts were the same. For the 1 mL/kg Carica papaya group, aspirin-induced and day 7 post-treatment platelet counts were the same. However, day 7 post-treatment platelet count was significantly lower compared with day 14 post-treatment by 103,862.2 plt/μL. The distilled water group also showed a significantly lower aspirin-induced platelet count compared with day 7 post-treatment by 128,221.2 plt/μL. However, day 7 and day 14 post-treatment platelet counts were the same. One-way ANOVA of day 7 post-treatment platelet counts showed that there was no significant difference between the three groups, indicated by a p value of 0.535. Day 14 post-treatment platelet counts also did not show a significant difference across treatment groups, shown by a p value of 0.341.


In conclusion, since the control and experimental groups did not exhibit a significant difference after seed extract administration, it can be inferred that Carica papaya seed extract was not an effective anti-thrombocytopenic agent during 7th and 14th day post-treatment. The researchers recommend that the time intervals between extractions and platelet counts be shortened to under a week, in order to properly determine whether the increase in platelet count is due to the Carica papaya seed extract or merely the body's natural mechanism of replenishing platelets.
