Raw soybean (Glycine max): An alternative to nutrient agar as enrichment medium for Proteus species and Streptococcus pneumoniae.

     This study determined the effectiveness of Raw Soybean Agar as an alternative enrichment medium to Nutrient Agar for Proteus species and Streptococcus pneumoniae. The researchers utilized a randomized post-test only experimental control group design. A total of forty (40) petri dishes were used in the study, twenty plates were that of Raw Soybean Agar and another twenty (20) for Nutrient Agar.Bacterial suspensions were prepared from the pure culture of the test organisms and was compared with that of zero point five (0.5) McFarland Standard to standardize the amount of bacteria present in the suspension. Pour Plate Method was performed to quantitate colonies that grew on the medium using a one is to twenty (1:20) dilution. Colonies on the medium were counted using a Quebec colony counter. Gram staining was performed to identify the morphology of the bacteria. Biochemical testing for Proteus species and Bile Solubility Test for Streptococcus pneumoniae were performed to confirm these test organisms.
      Results revealed comparable growth between the Raw Soybean Agar and the Nutrient Agar. Results showed that there was no significant difference in the colony count. Gram staining results revealed Gram-positive diplococci for Streptococcus pneumoniae and Gram-negative coccobacilli for the Proteus species. Bile Solubility Test confirmed that the colonies found on the Raw Soybean Agar and Nutrient Agar were Streptococcus pneumoniae. IMVIC results plus urease test confirmed that the colonies found on both the Nutrient Agar and Raw Soybean Agar was Proteus species.
     Statistical treatment of the data revealed that the number of colonies that grew on Raw Soybean Agar showed comparable results with that of the Nutrient agar, making it an effective alternative to Nutrient Agar as enrichment medium for Proteus species and Streptococcus pneumoniae. The researchers would like to recommend a clearing agent for easier visualization of colonies in Raw Soybean Agar due to the presence of particles in the agar. The researchers also recommend a pH regulator that will not interfere with the colony growth to produce a neutral pH in the medium. For future researchers, since pigmentation was produced when Streptococcus pneumoniae was cultured on Raw Soybean Agar, they may study on what could have caused it as well.
