Red cabbage (Brassica oleracea var. capitata f. rubra) leaf extract as staining agent for Entamoeba, Ascaris and Trichuris species.

The study aimed to determine the potential of Brassica oleracea var. capitata f. rubra (red cabbage) leaf extract as staining agent for Entamoeaba, Ascaris and Trichuris species. It utilized a repeated measures design. One-sample comparison of proportions was used for the basis of sample size computation, wherein forty-four (44) slides per group per specie (Entamoeba, Ascaris and Trichuris) were utilized. Stool specimens positive for Entamoeba, Ascaris and Trichuris species stained with red cabbage was used as the experimental group and same species were stained with Lugol's iodine as positive control group. Forty-four (44) slides stained with red cabbage and forty-four (44) slides were stained with Lugol's iodine.

All forty-four (44) slides per specie that presented with Entamoeba, Ascaris and Trichuris species stained positively with Lugol's iodine. None stained with the red cabbage leaf extract for the experimental group. However, the outer wall of Ascaris species demonstrated faint reddish purple and Trichuris was demonstrated with slightly reddish purple on its inner wall of the barrel shape when fine adjustment knob was used. Significant difference was not computed because all forty-four (44) slides stained negative with the red cabbage extract.


Based on the results of the study, Brassica oleracea var. capitata f. rubra (red cabbage) leaf extract was not able to stain the Entamoeba, Ascaris and Trichuris species. The researchers would like to recommend using another extraction process like the use of soxhlet and rotavap apparatuses.  And also the use of an alcohol-based solvent such as ethanol or methanol as extracting agent.  Moreover, it is highly recommended that another microscopic organism, such as trematodes be used as the subject of the study.
